RTP: Is it compulsory?

When you finally make your game, and build an EXE of it and then distribute it. If the people who download it don't have the RTP/RMXP installed, then the game won't run. Is there a way to make the game run and not having the need for the user to install the RTP/RMXP?

You'll have to import all the resources you use, so that it has no dependency on the RTP itself at all. If you're not sure which ones you've used and which ones you haven't, just import all the RTP files.

Then go to Game > Select RTP, and set all three of them to none.

Actually, the "RTP" refers to "RGSS-RTP Standard".  It is theoretically possible to create new RTPs by creating a folder under Program Files/Common Files/Enterbrain/RGSS/  and setting the appropriate registry key (see the Help file topic "RGSS Specifications"), but I have never seen any other RTPs than "Standard" or "RPGVX".

A single game can use up to three RTPs, two of them would have to be other than "Standard".
