Making NPC disappear?

Hello, everyone,
I've got the following problem: I'm currenty making my intro. I have two NPC talking to each other. One of them leaves to room, aka: walks up some stairs and should disappear. And that's my problem: How can I make an NPC seemingly coming from another room or disappearing into another room?
I just started today and all I know about RPG Maker XP I learned from Willow Sidhe over at Youtube, so, I beg you, please use easy words to explain.:*)

Just a small additional question:
If I wish an NPC to appear out of nowhere, should I set an Event, make a "Set Move Route", set it to "Graphic None", and, set it again, later, to "Graphic [insert desired graphic here]"?

That's one option, though as a result you'd start out with a npc, then have them disappear, then reappear (If I'm understanding you correctly.) I'll show you how I handle appearing and disappear NPCs (well, this example is actually a party member, but same idea.)

First I start with a blank event. In this case, because they will be "steping out of" the player sprite, the event is set on through.

I'll often have the blank event tucked away in a corner and then use "Set Event Location" before setting the Graphic.
